Yes. My cousin has often pointed out such instances even in the cassette versions. In 'Ayiram udhai vAngiya abUrva sigAmaNi' for instance, vElUr pattAbi (the politician S. Ve. Shekar) says, "eththanai kELvi mansukkuLLa irukkudhu, kEkka mAttenguRAm pAththiyA indhALu!" This is probably because the co-actor forgot to ask that question. What's remarkable is how the unscripted lines are just as funny if not more. I heard a blogger give another anecdote some time back. In 'alwA,' one of the actors was supposed to faint on stage, but while he fell on the floor, his vEtti malfunctioned a little bit. And SVS goes, "vizhaRadhu dhAn vizhaRa, ippidi AbAsamA vizhaNuma?"
